Stripping

Strip the signal from the LM or from the previous sequencing cycle with 100% Formamide. This step takes about 10 minutes

Blocking

This step is only carried out during the first cycle of sequencing. This step takes about 10 minutes.

Autofluorescence quenching (optional)

This step is only carried out when tissue samples present high autofluorescent background. This step takes about 10 minutes.

Hybridizing and Sequencing

Adapter probes and sequencing mix are hybridized sequentially. This step takes about 1.5 hours.

Imaging

It is recommended to work in an RNase free environment to avoid degradation of RNA molecules. Always use fresh RNAse free reagents for samples preparation.
